Metab. 39): E1112–E1118, 1999.—The distributions of the mRNAs for estrogen receptors (ERa and ERb) and their binding properties in myometria of pregnant and nonpregnant women and in leiomyoma were studied. RT-PCR analysis indicated that the term pregnancy myometria had little ERa mRNA, whereas the amounts of ERb mRNAs in pregnant or nonpregnant myometria appeared to be similar. Both ERa and ERb mRNA were greater in certain leiomyoma than in normal nonpregnant myometria. The binding kinetics revealed that two specific binding sites (with high or low affinity) for 17b-estradiol were present in the nonpregnant myometrium. Only the low-affinity binding sites were detectable in late-pregnancy myometria and in leiomyoma, and their capacities were increased twoto threefold (P , 0.001) in leiomyoma. The pregnancyand leiomyoma-related changes in myometrial ER status, especially the low concentration of ERa mRNA and the lack of high-affinity ER in pregnant women, plus the increased ERa and ERb mRNAs and the increased low-affinity ER in some leiomyoma, suggest that the redistribution of ER subtypes is associated with the pathological and/or normal growth of the myometrium.

Augmented endothelial nitric oxide synthase (eNOS) protein expression in human pregnant myometrium: possible involvement of eNOS promoter activation by estrogen via both estrogen receptor (ER)a and ERb

The aim of the present study was to investigate the possible contribution of estrogen to pregnancy-associated modulation of nitric oxide production in the human myometrium during pregnancy. Both endothelial nitric oxide synthase (eNOS) and inducible NOS (iNOS) proteins were clearly expressed in the non-pregnant myometrium and were elevated in the ®rst trimester of pregnancy. Oral contraceptive ...

Estrogen Receptor, Progesterone Receptor and Glucocorticoid Receptor Expression in Normal Myometrium and in Leomyoma

Uterine leiomyoma is the most common benign smooth muscle tumour that originates from the myometrium. Ovarian sex steroids are responsible for the growth of such tumours, though estrogen and progesterone are not the only modulators involved in the tumourigenesis and growth of leiomyomata. The activation of glucocorticoid receptor (GR) can induce the expression and activity of estrogen sulfotran...

Potential link between estrogen receptor-alpha gene hypomethylation and uterine fibroid formation.

Uterine leiomyomas are the most common uterine tumors in women. Estrogen receptor-alpha (ER-alpha) is more highly expressed in uterine leiomyomas than in normal myometrium, suggesting a link between uterine leiomyomas and ER-alpha expression. DNA methylation is an epigenetic mechanism of gene regulation and plays important roles in normal embryonic development and in disease progression includi...
